Figure 1: Uptake of fluorescein diacetate in thymocytes as a function of interaction time. Thymocyte cell suspension prepared in RPMI 1640 was incubated with fluorescein diacetate (final concentration 10 μg/ml) at 4°C. Aliquots taken after increasing time of incubation were washed with ice cold phosphate-buffered saline by centrifugation at 2000 rpm at 4°C and fluorescence intensity of 100 μl cell suspension was measured after dilution in 3 ml phosphate-buffered saline at 25°C
